Toward PRecisiOn Medicine for the Prediction of Treatment Response in Major Depressive Disorder through Stratification of Combined Clinical and Omics Signatures

Major depressive disorder (MDD) is the most common psychiatric disorder worldwide. Pharmacotherapy is the standard first-line treatment, yet only one third of patients respond to the initial trial and about 30% develop treatment-resistant depression (TRD), which is associated with specific clinical and molecular features. The PROMPT project aims to develop a precision medicine algorithm for early identification of non-responders. The study is organized in two phases: Phase 1 includes a retrospective cohort of 300 MDD patients (150 TRD and 150 responders) and Phase 2 follows a naturalistic cohort of MDD patients to validate the predictive performance of the algorithm under real-world conditions. Both phases include deep clinical and molecular profiling (genomic, transcriptomic and miRNomic data from both phases) of MDD patients.
