A new non-invasive prenatal paternity test based on double nucleotide polymorphisms abates sequencing uncertainties and improves sensitivity
This study presents the development and validation of NEWPAT, a non-invasive prenatal paternity testing (NIPAT) approach based on fetal cell-free DNA (cfDNA) and Double Nucleotide Polymorphisms (DNPs). A targeted amplicon-based NGS panel including 979 markers was applied to genomic DNA, simulated maternal-fetal mixtures, and cfDNA samples from pregnant women and putative fathers. High-depth sequencing data were generated and analyzed to evaluate DNP performance for fetal DNA detection and paternity inference.
