In vitro depletion of Caveolin-1 in human plasmablast cells
In humans, unlike in mice, Cav-1 is expressed not only after B-cell activation but also in PC progenitors, plasmablasts, and mature PCs. To assess its functional role during B-cell differentiation, we used our previously published in vitro B-cell differentiation model and transfected day-5 cells with CAV1-targeting siRNAs. This resulted in a significant reduction of both CAV1 mRNA and protein levels by day 7, compared with scrambled siRNA controls. Bulk RNA-seq was then performed on day-7 plasmablasts transfected with siRNA-CAV1 versus siRNA control to compare their transcriptomic profiles (n = 4 experiments).
