Effect of TNFR2 agonism on CD4+ T cells (single-cell level) from healthy and SLE donors

We treated peripheral blood mononuclear cells from healthy donors and systemic lupus erythematosus patients with (i) a human-specific tetravalent VHH-Fc fusion TNFR2 agonist, (ii) isotope control VHH, (iii) an IL2-mutein and (iv) no treatment for 2 days at 4°C. CD4+ T cells were analyzed by scRNA/TCR-seq, including surface protein measurement (CITE-seq) for CD120b.
