TIM3 blockade with hypomethylating therapy restores NK and cytotoxic CD4+ T cell activity in patients with AML or MDS
Acute myeloid leukemia (AML) remains a therapeutic challenge, requiring novel combination strategies to improve patient outcomes. This study investigates the cellular and molecular dynamics of bone marrow microenvironments in patients receiving combination therapy with anti-TIM3 and decitabine within a phase Ib clinical trial (NCT03066648). By analyzing longitudinal samples, the project aims to characterize treatment response, resistance mechanisms, and microenvironmental shifts at single-cell resolution. The resulting insights provide a comprehensive framework for evaluating checkpoint inhibition combined with hypomethylating agent in AML and MDS.
