Whole-genome sequences of the Korean population generated using long-read technologies

We performed long-read whole-genome sequencing of 19 Korean individuals to construct a Korean pangenome reference. Genomic DNA was extracted from peripheral blood of 16 individuals and from lymphoblastoid cell lines (LCLs) of 3 individuals, and DNA quality and quantity were assessed prior to library preparation. Twelve individuals underwent HiFi long-read sequencing on the PacBio Revio platform (mean raw depth ~26×), and seven individuals underwent long-read sequencing on the Oxford Nanopore PromethION platform (mean raw depth ~24×). Two of the Revio-sequenced individuals (KOR-003 and KOR-004) additionally underwent Hi-C sequencing (Dovetail Omni-C, Illumina NovaSeq X) to support haplotype-resolved, chromosome-scale assembly, and KOR-003 was also sequenced on the PromethION platform. Samples are identified as KOR-003 to KOR-021 (recorded as KOR_003 to KOR_021 in the sample aliases). Whole-genome sequences of three further Korean individuals, generated previously using the PacBio Sequel II system, are available in the European Genome-phenome Archive (EGA) under study accession EGAS50000000375.
