Single-cell profiling reveals conserved epithelial composition and CFTR expression in human nasal epithelial and BCi airway cultures.
We generated single-cell RNA-sequencing profiles from differentiated human nasal epithelial (HNE) cells obtained from one healthy donor and from the immortalized human airway basal cell line BCi-NS1.1. Both models were differentiated under air-liquid interface conditions for 30 days before cell dissociation, single-cell partitioning, barcoding, and sequencing using the 10x Genomics Chromium Single Cell 3′ Reagent Kit v4. The dataset contains approximately 30,146 cells and was generated to compare epithelial cell composition and CFTR expression between primary HNE-derived and BCi-derived airway epithelia. The data identify basal, suprabasal, ciliated, deuterosomal, secretory, goblet, ionocyte, macroalveolar, and neuroendocrine populations and support evaluation of BCi-NS1.1 as a complementary model for cystic fibrosis airway research.
